Most published studies on thymosin alpha-1 report changes in immune measurements rather than clinical outcomes, and findings differ across designs and populations. Whether the peptide signals through one defined receptor or through several less specific interactions remains an open question. Its reported circulation half-life of a few hours complicates comparison of dosing schedules across trials. Mechanistic claims are frequently drawn from isolated cell cultures, and how far those results extend to whole organisms is unresolved.
Thymosin alpha-1 is a synthetic peptide of 28 amino acids whose sequence matches the amino-terminal region of prothymosin alpha. The chain is acetylated at its first residue and contains one disulfide bridge between two cysteine residues, which folds the molecule into a compact loop. Its molecular formula, C129H215N33O55, corresponds to a monoisotopic mass of roughly 3,106 daltons. Material used in laboratories is made by solid-phase synthesis rather than isolated from animal tissue.
Early work on thymic extracts in the 1960s described a heat-stable acidic fraction containing many polypeptides. Separation of that mixture yielded individual components, and thymosin alpha-1 was named as one of them on the basis of assays for T-cell activity. The first preparations came from calf thymus, while subsequent research and clinical material has been chemically synthesized. Nomenclature in older papers is inconsistent, and the same peptide sometimes appears under different designations, which complicates literature searches.

The molecule consists of 28 amino acid residues with an acetyl group attached to the N-terminal serine. Its sequence is acidic overall, with several glutamic and aspartic acid residues distributed along the chain and no cysteine, so disulfide bridges do not form. The peptide carries a net negative charge at physiological pH. Because the N-terminus is blocked, the intact molecule resists degradation by many aminopeptidases, which contributes to its stability in biological fluids.
The peptide is generated in cells by cleavage of prothymosin alpha, a larger acidic protein encoded by the PTMA gene. Prothymosin alpha is expressed in many tissues, not only in the thymus, and its functions include nuclear roles in chromatin-related processes. The 28-residue fragment corresponds to the N-terminal portion of that precursor. How the cleavage occurs and how the fragment's concentration is regulated remain open questions; circulating amounts are small and difficult to measure reliably with routine assays.
Thymosin alpha 1 is a short peptide first isolated from bovine thymus tissue in the early 1970s during fractionation work aimed at identifying factors that influence T cell development. It belongs to a family of acidic thymic peptides, and the original preparations contained several components that were later separated by chromatography. The compound is now produced synthetically rather than extracted from tissue, which removes batch variability tied to animal sourcing. Researchers describe it as an immunomodulatory peptide because laboratory studies show effects on several cell types of the innate and adaptive immune systems.
Laboratory supplies of the peptide usually arrive as a lyophilized powder in sealed vials. The powder is hygroscopic, so a vial should be allowed to reach room temperature before it is opened to prevent condensation on the contents. Weighing and transfer are best performed in a low-humidity environment with clean tools. Once dissolved, the solution should be mixed gently rather than vortexed, because foaming and shear can reduce recovery of the peptide.
Storage recommendations center on low temperature, dryness, and protection from repeated freezing and thawing. The intact powder is commonly held at 20 degrees below zero Celsius or colder, while a working solution is divided into single-use aliquots to limit freeze-thaw cycles. Buffered saline or phosphate-buffered saline at neutral pH is frequently used as a diluent. Light sensitivity is not well documented, yet amber vials or foil wrapping are common practice for long-term storage of peptide stocks.
A major focus of pharmacometrics is to understand variability in drug response. Variability may be predictable (e.g., due to differences in body weight or kidney function) or apparently unpredictable (reflecting the current knowledge gap).
=== Cosmetic surgery === Videos about cosmetic surgery are very popular on TikTok. In January 2022, videos with hashtags related to plastic surgery had over 29 billion views combined on the platform. TikTok and Instagram have led to an increase in the number of cosmetic surgeries performed on young people. In 2021, Plastic and Reconstructive Surgery published an article that found that plastic surgeons were among the earliest adopters of social media and at the time the article was published, it was found that at least five plastic surgeons had surpassed 1 million followers on TikTok. The article noted that some surgeons were influencers on the platform and had the ability to influence public perception. A 2021 study published by the University of South Florida found that content posted on TikTok by plastic surgeons helped legitimize plastic surgery by educating their viewers and reducing their fear of the surgeries. Plastic surgery is also legitimized by the TikTok recommendation system which shows users who showed interest in plastic surgery videos even more plastic surgery videos via the For You Page, which makes plastic surgery seem more widespread than it actually is among both celebrities and normal people. TikTok does not allow direct paid advertisements of cosmetic surgeries on its platform, but cosmetic surgery clinics are able to promote their services using normal unpaid posts, as well as by paying influencers or giving them free surgeries in exchange for the influencer posting a video about their cosmetic surgery experience.
=== Elution-extrusion === The extrusion of stationary phase from the column at the end of a separation experiment by stopping rotation and pumping solvent or gas through the column was used by CCC practitioners before the term EECCC was suggested. In elution-extrusion mode (EECCC), The mobile phase is extruded after a certain point by switching the phase being pumped into the system whilst maintaining rotation. For example, if the separation has been initiated with the aqueous phase as the mobile phase at a certain point the organic phase is pumped through the column which effectively pushes out both phases that are present in the column at the time of switching. The complete sample is eluted in the order of polarity (either normal or reversed) without loss of resolution by diffusion. It requires only one column volume of solvent phase and leaves the column full of fresh stationary phase for the subsequent separation.
=== Lyophilized === Lyophilized drugs are stored in vials, cartridges, dual chamber syringes, and prefilled mixing systems. Lyophilization, or freeze drying, is a process that removes water from a liquid drug creating a solid powder, or cake. The lyophilized product is stable for extended periods of time and could allow storage at higher temperatures. In protein formulations, stabilizers are added to replace the water and preserve the structure of the molecule. Before administration, a lyophilized drug is reconstituted as a liquid before being administered. This is done by combining a liquid diluent with the freeze-dried powder, mixing, then injecting. Reconstitution usually requires a reconstitution and delivery system to ensure that the drug is correctly mixed and administered.
A protein microarray (or protein chip) is a high-throughput method used to track the interactions and activities of proteins, and to determine their function, and determining function on a large scale. Its main advantage lies in the fact that large numbers of proteins can be tracked in parallel. The chip consists of a support surface such as a glass slide, nitrocellulose membrane, bead, or microtitre plate, to which an array of capture proteins is bound. Probe molecules, typically labeled with a fluorescent dye, are added to the array. Any reaction between the probe and the immobilised protein emits a fluorescent signal that is read by a laser scanner. Protein microarrays are rapid, automated, economical, and highly sensitive, consuming small quantities of samples and reagents. The concept and methodology of protein microarrays was first introduced and illustrated in antibody microarrays (also referred to as antibody matrix) in 1983 in a scientific publication and a series of patents. The high-throughput technology behind the protein microarray was relatively easy to develop since it is based on the technology developed for DNA microarrays, which have become the most widely used microarrays.

==== Bone and collagen ==== Cortisol reduces bone formation, favoring long-term development of osteoporosis (progressive bone disease). The mechanism behind this is two-fold: cortisol stimulates the production of RANKL by osteoblasts which stimulates, through binding to RANK receptors, the activity of osteoclasts, cells responsible for calcium resorption from bone, and also inhibits the production of osteoprotegerin (OPG) which acts as a decoy receptor and captures some RANKL before it can activate the osteoclasts through RANK. In other words, when RANKL binds to OPG, no response occurs as opposed to the binding to RANK which leads to the activation of osteoclasts. It transports potassium out of cells in exchange for an equal number of sodium ions (see above). This can trigger the hyperkalemia of metabolic shock from surgery. Cortisol also reduces calcium absorption in the intestine. Cortisol down-regulates the synthesis of collagen.
== Prognosis == With treatment, the five-year survival rate is >80%, and fewer than 30% of affected individuals require long-term dialysis. A study performed in Australia and New Zealand demonstrated that in patients requiring renal replacement therapy (including dialysis), the median survival time is 5.93 years. Without treatment, virtually every affected person will die from either advanced kidney failure or lung hemorrhages.
Berkelium is a synthetic chemical element; it has symbol Bk and atomic number 97. It is a member of the actinide and transuranium element series. It is named after the city of Berkeley, California, the location of the Lawrence Berkeley National Laboratory (then the University of California Radiation Laboratory) where it was discovered in December 1949. Berkelium was the fifth transuranium element discovered after neptunium, plutonium, curium and americium. The major isotope of berkelium, 249Bk, is synthesized in minute quantities in dedicated high-flux nuclear reactors, mainly at the Oak Ridge National Laboratory in Tennessee, United States, and at the Research Institute of Atomic Reactors in Dimitrovgrad, Russia. The longest-lived and second-most important isotope, 247Bk, can be synthesized via irradiation of 244Cm with high-energy alpha particles. Just over one gram of berkelium has been produced in the United States since 1967. There is no practical application of berkelium outside scientific research which is mostly directed at the synthesis of heavier transuranium elements and superheavy elements. A 22-milligram batch of berkelium-249 was prepared during a 250-day irradiation period and then purified for a further 90 days at Oak Ridge in 2009. This sample was used to synthesize the new element tennessine for the first time in 2009 at the Joint Institute for Nuclear Research, Russia, after it was bombarded with calcium-48 ions for 150 days. This was the culmination of the Russia–US collaboration on the synthesis of the heaviest elements on the periodic table.
==== Dangers ==== If taken undiluted by mouth, GBL can cause esophageal and gastro-intestinal irritation. It is possible for oral ingestion of GBL to cause nausea and other similar problems, possibly more so than with GHB. GHB has biphasic effects, a euphoric effect at low doses (the reason for the term liquid ecstasy), and a sedative effect at higher doses. As a result of this sedation it can cause unconsciousness. When combined with alcohol the increased sedation and risk of vomiting results in a high risk of fatality. As a result, many harm reduction organizations suggest never mixing the two drugs. There have been several news reports of deaths associated with GBL, usually in combination with alcohol or other depressants.

=== Acute === Acute poisoning is exposure to a poison on one occasion or during a short period of time. Symptoms develop in close relation to the exposure. Absorption of a poison is necessary for systemic poisoning. Furthermore, many common household medications are not labeled with skull and crossbones, although they can cause severe illness or even death. Poisoning can be caused by excessive consumption of generally safe substances, as in the case of water intoxication. Agents that act on the nervous system can paralyze in seconds or less, and include both biologically derived neurotoxins and so-called nerve gases, which may be synthesized for warfare or industry. Inhaled or ingested cyanide, used as a method of execution in gas chambers, or as a suicide method, almost instantly starves the body of energy by inhibiting the enzymes in mitochondria that make ATP. Intravenous injection of an unnaturally high concentration of potassium chloride, such as in the execution of prisoners in parts of the United States, quickly stops the heart by eliminating the cell potential necessary for muscle contraction. Most biocides, including pesticides, are created to act as acute poisons to target organisms, although acute or less observable chronic poisoning can also occur in non-target organisms (secondary poisoning), including the humans who apply the biocides and other beneficial organisms. For example, the herbicide 2,4-D imitates the action of a plant hormone, which makes its lethal toxicity specific to plants. Indeed, 2,4-D is not a poison, but classified as "harmful" (EU).
== Function == As well as C. maenas, CCAP has cardioacceleratory properties in Manduca sexta and Drosophila melanogaster; it is a highly conserved protein found in species as diverse as moths and fruit flies in addition to the crustacean it was originally identified in. Under specific conditions, CCAP has also been shown to induce cardiac reversal in M. sexta and D. melanogaster. Since CCAP was first identified and its possible role suggested, it has been proven it serves many other functions. It regulates release of adipokinetic hormone during sustained flight in L. migratoria. It is also important in metabolism and digestion in other species; CCAP is involved as a hormone in the cockroach Periplaneta americana by upregulating digestive hormones after the insect has fed on nutritional matter. In the crab Cancer borealis CCAP controls passage of food through the foregut by modulating the stomatogastric ganglion's pyloric rhythm. This suggests CCAP is involved primarily in movement of food along the gut, facilitating digestion by secretion of digestive hormones and increasing flow of blood. The effects of CCAP on the reproductive organs of females of the species L. migratoria have also been studied. The peptide stimulates the contraction of the oviducts and spermatheca. There is no CCAP or CCAP-like immunoreactivity shown in the oviduct or the nerves innervating the oviduct so it is probable that CCAP acts as a neurohormone to perform these functions. CCAP is one of several peptides and hormones to be involved in the moulting process of crustaceans and insects.
The lunula is the crescent-shape area at the base of the nail, lighter in color as it mixes with matrix cells. Only primates have nails. In other vertebrates, the keratinizing system at the terminus of each digit produces claws or hooves. The epidermis of vertebrates is surrounded by two kinds of coverings, which are produced by the epidermis itself. In fish and aquatic amphibians, it is a thin mucus layer that is constantly being replaced. In terrestrial vertebrates, it is the stratum corneum (dead keratinized cells). The epidermis is, to some degree, glandular in all vertebrates, but more so in fish and amphibians. Multicellular epidermal glands penetrate the dermis, where they are surrounded by blood capillaries that provide nutrients and, in the case of endocrine glands, transport their products.

Its sequence corresponds to the amino-terminal portion of prothymosin alpha, a larger protein present in many cell types. The isolated 28-residue peptide is a fragment of that protein rather than a separately encoded molecule, and laboratory material is produced by synthesis.
The single bridge between two cysteine residues holds the chain in a folded loop that influences its shape and its behavior in solution. Loss of the bridge through reduction or oxidation shifts chromatographic retention and is tracked during stability work.
It is a defined 28-residue sequence derived from a larger precursor, whereas many other thymic preparations are mixtures of several polypeptides. Its acetylated amino terminus and single disulfide bridge distinguish it chemically from unrelated thymic extracts.
