== Diagnosis == Initially diagnosis is typically established by creatine, creatinine, and guanidinoacetate measurement in the plasma, urine, and/or cerebrospinal fluid, as listed in the table below. The levels of these biochemical markers can indicate which specific creatine disorder is present. Brain magnetic resonance spectroscopy can also be used in diagnosis, but will show decreased creatine levels in all three disorders and is therefore unable to identify the specific creatine deficiency disorder. Definitive diagnosis can be found through DNA sequencing of the relevant gene and enzymatic activity (for GAMT and AGAT) or transport activity (CTD).
=== Eyes === Isotretinoin and other retinoids are well known to affect the eyes. Dry eyes are very common during treatment and is caused by isotretinoin's apoptotic effect on the meibomian glands. Some people develop contact lens intolerance as a result. In some people, these changes are long-lasting or irreversible and represent Meibomian Gland Dysfunction (MGD). Other common effects on the eyes include inflammation of the eyelid (blepharitis), red eye caused by conjunctivitis and irritation of the eye. More rare ocular side effects include blurred vision, decreased night vision (which may be permanent), colour blindness, development of corneal opacities, inflammation of the cornea (keratitis), swelling of the optic disk (papilloedema, associated with IIH), photophobia and other visual disturbances.

=== Wounds === In wounds, CGRP receptors found in nerve cells deactivate the immune system, to prevent collateral damage in case of a clean wound (common case). In very preliminary research, nerve blockers like lidocaine or botox have been demonstrated to block CGRP cascade, thereby allowing immune system involvement and control of pathogens, resulting in complete control and recovery.
